Why DEPC-Free Matters

DEPC (diethylpyrocarbonate) is the most common chemical treatment for nuclease removal — but DEPC residue is a documented inhibitor of downstream enzyme reactions, especially reverse transcription and in vitro translation. Most autoclave protocols are designed to convert DEPC to ethanol and CO₂, but trace residue is a recurring debate in molecular biology literature.
